Reserve a full day for a trip into the Mekong. Here's a tip to save you some bucks - take a 5 minute cab from District One to the backpackers area (Pham Ngu Lao) at around 730 or 800am and sign up for tour from there (or go the night before). Same bus - same guide - same everything... but at a fraction of the price of booking at your hotel. The tour I went on bused us to the Mekong where we jumped on a boat into the mangroves which were used by the VC to escape from American patrols during the war. Quite a fascinating trip and worth the bus ride.
